You can take the U-Bahn from the airport to the Stadtmitte station and transfer there to a train to the Spittelmarkt station. The hotel will be about 300 m from the stop if the address you listed is correct. It takes about 30 minutes on the U-bahn and another couple minutes walk.

The hotel is located directly opposite the subway station “Märkisches Museum”
There is no U-Bahn at the Berlin-Tegel airport, the other Y!A contributor may have mixed it up with the Berlin International Airport at Tempelhof.
From Tegel you take Bus X9 to Ernst-Reuter Platz (16 minutes) and change to subway U2 in the direction of Pankow (19 minutes). Märkisches Museum is the 12th stop. The stop before is called Spittelmarkt. Cost for a single ticket (Zone AB) is 2,10 €.http://www.fahrinfo-berlin.de/Fahrinfo/b…
